can you use partitions on USB drives?

I have a 500Gb desktop hard-drive thats formatted NTFS for my TV so I can watch 1080p MKV files through the USB port on my LG LH3000 1080p TV

- if I was to partition a 100Gb off for the Wii and formatted that partition with WBFS would it work? - as in read that partition? or does the whole drive gotta be done?

OK cheers - will give it a go - problem is the drives almost hlaf full with HD files I cant store anywhere if had to format so its GOT to be done with the files on the drive - it is possible just may take some time -
